Genetic divergence and relationships among Atractylodes spp. by DNA barcode system
2011
Atractylodes spp.(Baekchul) has high demand next to Glycyrrhiza spp.(Gamcho) in industry of the dietary supplements and oriental herbal medicine. However, Atractylodes is depends on imports because its output and supply are scarce. It has possibility to mix with imported products in the domestic. Thus, it is required to develop the origin discrimination method using molecular marker besides morphological discrimination for preventing the mixed with the domestic and imported products. This research was accomplished for genetic divergence comparison through sequence analysis which used chloroplast and nuclear DNA barcode system among Atractylodes spp.(A. japonica, A. macrocephala and A. hybrid Dachul). As a result, chloroplast psbA-trnH and rpoB were considered possibly with barcode DNA because the SNP region was confirmed, whereas rpoC1 was unsuitable for barcode DNA. Using of ribosomal ITS2 could confirm the SNP region about 50, and we will accomplish reconfirmation of this result. Also, we have a plan to accomplish PCR analysis and sequencing of the extracted gDNA from dry medicinal material. ---------------------------------------------------Corresponding author: Tel. 02-2260-3308, e-mail: jhcho@dongguk.edu
