Eukaryotic expression and immunogenicity detection of Newcastle disease virus(NDV)F gene

To assess the probability of F gene DNA vaccine in controlling ND,the NDV F gene was inserted into eukaryotic expression vector pcDNA3.1/V5-His-TOPO,and an eukaryotic expression recombinant plasmid pcDNA NDV ZF,which containing start codon and stop codon,was constructed.The recombinant plasmids were transfected into CEF cells with the liposome,and the F protein was detected by indirect immunofluorescence experiment.The results demonstrated that F protein could be expressed in a large amount in CEF cells.The pcDNA NDV ZF was injected into muscles of 1-d-old SPF chickens at a dose of 100 μg.Two weeks post injection,antibody to NDV in serum of the immunized chickens were detected with indirect ELISA.The results showed that the NDV F gene DNA vaccine plasmids could induce the specific antibodies in chickens and had good immunogenicity.
