Culture and identification of mouse osteoblasts by modified enzymatic digestion in vitro

【Objective】To optimize the method to isolate primary osteoblasts from new born mice efficiently. 【Methods】Osteoblasts were obtained cleanly by digestion of collagense A and dispase from 72h new born mice. Typical morphological and biological characteristics of the osteoblasts were identificated by observation under optic microscope, assay of Alkaline phosphatase (ALP) activity, ALP staining, Von Kossa staining and Alizarin Red staining. 【Results】The results showed that the isolated cells had typical morphological and biological characteristics of the osteoblasts.【Conclusion】The modified enzymatic digestion is advantageous and efficient without trypsinase and shearing cranial bone.
