The pharmacological study of compound piracetam and galantamine on Alzheimer's disease

2011
OBJECTIVE To investigate the effect of Compound piracetam on Alzheimer's disease rats, and compared with piracetam or galantamine use alone.METHODS Seventy normal Wistar rats(male 35,female 35) were randomly divided into seven groups(n=10)including sham-operated,model,high dose compound piracetam, medium dose compound piracetam, low dose compound piracetam, piracetam and galantamine groups.except the sham-operated and model groups were administrated with distilled water, other groups were administrated with different dose medicine after the second day of the operation. The abilities of learning and memory of rats were investigated by step-down test on the 9th day after the operation, the changes of the pathology and the activity of the Ach in hippocampus were measured on the 14th day after the operation.RESULTS Compared with model and single component group, the learning and memory abilities were significantly improved(P0.05),and the brain acetylcho content was increased(P0.01) in high and medium dose compound piracetam.the granulose cell of hippocampus in high and medium dose compound groups was lined up in order and no abnormal change was found.CONCLUSIONCompound piracetam can improve the therapeutic effect on AD superior to the single component.
