Study on the methylation level of FOXP3gene promoter region in human hepatoma cell lines

[Objective]To detect the methylation level of transcription factor forkhead box protein 3(FOXP3)gene promoter regions in human hepatoma cell lines.[Methods]Target DNA was extracted from the human hepatoma cell lines HepG2 and SMMC7721and treated with bisulfate sodium.Clone sequencing was then used to detect methylation of the FOXP3 gene promoter regions.[Results]The promoter CpG island of FOXP3 in the human hepatoma cell lines HepG2 showed 86% methylation frequency,while 80%in SMMC7721 cells.[Conclusion]The methylation frequency of FOXP3 promoter regions in human hepatoma cell lines has been successfully identified in a high level,which might lay some groundwork for further study about epigenetic research of FOXP3 in hepatocellular carcinoma(HCC).
