The effect of developing crystallin on survival and growth of rat RGCs in vitro

Objective To study whether crystallin of rats at different growth stages has different protective effects on survival and growth of rat retinal ganglion cells (RGCs) in vitro.Methods RGCs in SD rats were cultured in DMEM/F12 containing crystallin of different age rats(1 day,2 weeks,6 months and 1 year).The RGCs survival time,process growth and apoptosis were observed.Results The best concentration of soluble crystallin was 10-5g/L after comparing three different concentrations.Crystallin of different growth stage rats could prolong RGCs' survival time,which increased with the increase of growth period.However,1d group and 2w group did not differ significantly.RGCs' apoptosis decreased after crystallin was added and it gradually decreased with prolonged growth period.Conclusion Crystallin can significantly promote the survival and growth of rat RGCs in vitro.And crystallin of 10-5g/L is the most effective concentration among various concentrations.Crystallin of different growth stages has a different neuroprotective effect,which is enhanced with the extension of growth period.
