Study on genetic variation of Lolium multiflorum L. by RAPD markers

Genetic diversity and genetic variation of 40 accessions of Lolium multiflorum L.collected from Europe,America and China are analyzed using randomly amplified polymorphic DNA(RAPD) molecular markers with 18 RAPD primers.The following results are obtained.(1)307 discernible DNA fragments are generated with 291(78.65%) being polymorphic,the polymorphic information contents(PIC) ranged from 0.387(S3) to 0.499(S48) with an average of 0.478.(2) The genetic similarity(GS) among all accessions ranged from 0.59 to 0.93 with an average of 0.75,and genetic diversity are observed in abroad-cultivar(P,75.24%) and imported-cultivar(P,60.26%) groups.These results suggest that there is a poor genetic diversity among all Lolium multiflorum L.accessions tested.(3) Based on the cluster and principal component analysis,it suggests that RAPD markers can separate 40 accessions into cultivars and wild clearly,and the accessions from the same origin f requently clustered into one group.
