Objective: To explore the effects of ginsenoside-Rb1 on cell multiplication and [Ca(superscript 2+)](subscript i) in the SK-N-SH treated with chronic morphine. Methods: MTT was used to research the effects of ginsenoside-Rg1 on cell multiplication in the SK-N-SH cells treated with chronic morphine. Fluorospectrophotometry was used to observe the effects of ginsenoside-Rb1 on [Ca(superscript 2+)](subscript i) in the SK-N-SH cells treated with chronic morphine. Results: Morphine significantly inhibited cell multiplication and resulted in calcium overload in the SK-N-SH cells treated with chronic morphine for 48 h (P<0.01). Adding 16 μmol•L^(-1) ginsenoside-Rb1 could enhance cell multiplication (P<0.05). However ginsenoside-Rb1 (16 μmol•L^(-1), 32 μmol•L^(-1)) could relieve the depressant effect of morphine on cell multiplication (P<0.05). Adding 16 μmol•L^(-1) ginsenoside-Rb1 made [Ca(superscript 2+)](subscript i) significantly lower in SK-N-SH cells (P<0.01). Cell [Ca(superscript 2+)](subscript i) significantly increased (P<0.01) when stimulated by morphine. Ginsenoside-Rb1 (16 μmol•L^(-1), 32 μmol•L^(-1)) could significantly inhibit morphine induced calcium overload (P<0.01). Conclusion: Ginsenoside-Rb1 can significantly relieve the depressant effect of morphine on cell multiplication and elevate the morphine induced [Ca(superscript 2+)](subscript i) in the SK-N-SH cells.
