EFFECT OF ISCHEMIC POSTCONDITIONING ON KIDNEY ISCHEMIA REPERFUSION INJURY IN RABBITS

[Objective] To investigate the effect of ischemic postconditioning on kidney ischemia reperfusion injury and its mechanism in rabbits. [Methods] 18 Japanese white rabbits weighed 1.5-2.2kg were randomly allocated into 3 groups(n=6 in each group): sham operation group(S group);ischemia-reperfusion group(I /R group) and I /R + ischemic postconditioning group(IPo group) . All rabbits were anesthetized with intravenous 3% pentobarbital 1ml /kg. Rabbits were subjected right nephrectomy and 45-min left kidney occlusion. In S group,the blood flow was restored without intervention. In IPo group,45 min ischemia was followed by four cycles of 1 min episodes of ischemia and 1 min interval for reperfusion. The animals were killed at 60 min post reperfusion. Blood samples were taken from arteria cruralis for determination of blood urea nitrogen(BUN) and plasma creatinine(Cr) concentration and hemorheology. Apoptosis of the renal tubular epithelial cell was detected using TUNEL. [Results] Compared to S group,the concentrations of BUN and Cr and the number of apoptosis cells and the AI significantly increased(P﹤0.05) ,and hemorheology indexes were abnormal in the I /R group rabbits. Compared to I /R group,the concentrations of BUN and Cr and the number of apoptosis cells and the AI significantly decreased(P﹤0.05) and hemorheology indexes were improved in the IPo group rabbits. [Conclusion] Postconditioning could reduce ischemia reperfusion injury of rabbits’ kidneys through decreasing apoptosis of the renal tubular epithelial cells and improving hemorheology indexes.
