RFLP Standardization Report for DQ Beta/PvuII

Twenty-six bands were standardized with DQ beta/PvuII system as shown in Table 1. All bands present in Figure 1 were counted as standard bands since each band could clearly be assigned in more than one laboratory. Four bands–no. 4 (6.98kb), no. 20 (1.77kb), no. 22 (1.02kb), and no. 25 (0.53kb)–were nonpolymorphic. MacMurray et al.1 have previously reported three nonpolymorphic bands of 7.3 kb, 1.1 kb, and 0.7 kb obtained with the 5’ DQ beta probe which contains the untranslated region to the majority of the second domain. We consistently observed, however, both the 1.02 kb fragment and 1.27 kb fragment. Eight bands–no. 3 (8.19 kb), no. 4 (6.98 kb), no. 14 (3.72 kb), no. 19 (2.02 kb), no. 21 (1.66 kb), no. 23 (0.88 kb), no. 24 (0.63 kb), and no. 26 (0.34 kb)–showed no cross-hybridization with other beta probes (Fig. 2). The DQ beta cross- hybridizing bands are shown in Table 2 in the DR beta/PvuII report. Seven of 26 bands are correlated with specific DQw types. These bands are no. 19 (2.02 kb) and no. 18 (2.17 kb) with DQw6 subtypes and no. 24 (0.63 kb), no. 21 (1.66 kb) and no. 8 (4.78 kb) with DQw2 subtypes and no. 14 (3.72 kb), and no. 26 (0.34 kb) with DQw7.
