Correlation between Cyr61 and chronic myelogenous leukemia.

Objective To investigate the clinical significance of cysteine rich 61(Cyr61)in the bone marrow(BM)of chronic myelogenous leukemia(CML)patients.Methods Expressions of Cyr61 mRNA and Cyr61 protein were detected in bone marrow mononuclear cells(BMMNC)from 38 patients with CML(20 chronic phase,11 accelerated phase and 7 blast crisis)and 10 normal controls by the reverse transcriptase-polymerase chain reaction(RT-PCR)method and immunohistochemistry,respectively.Results Expression of Cyr61mRNA in a chronic phase,accelerated phase and blast crisis was significantly higher in CML patients than in normal controls(P0.05).Expression of the Cyr61 gene was higher in an accelerated phase and blast crisis than in the chronic phase(P0.05),but showed no significance(P0.05).Expression of Cyr61 protein was higher in CML patients than in normal controls(P0.05).Expression of Cyr61 protein was higher in the accelerated phase and blast crisis than in the chronic phase(P0.05).Conclusion Over-expression of the Cyr61 gene which plays an important role in the acute transformation of chronic myelogenous leukemia may relate to the progression and genesis of CML and provides an experimental basis for early diagnosis of CML blastic crisis.
