Adhesion assay for the fibroblast cells on the fibrin thin layer substrate

Sarcoma cells (S-180) and 3T3-SV-40 transformed cells were cultured on the fibrin thin layers to investigate the substrate dependence of the spreading activity, compared with those of the other non-malignant cells (3T3-, 3T6-fibroblasts, and baby hamster kidney cells). The cell spreading function of S-180 and 3T3-SV-40 transformed cells as well as the other cells was depressed by the crosslink-inhibition of fibrin and was lost on the non-crosslinked fibrin. This suggests that the inhibition of fibrin crosslinking could prevent the anchorage of the malignant fibroblast cells, and this adhesion assay would be a convenient method for the study on the substrate dependence of metastatic cells.
