Isolation and characterization of seed specific phytase promoter ( TaPAPhy_a1.1 ) from wheat

Phytase enzyme plays a vital role to hydrolyze well known anti-nutrient substance phytic acid which is found in wheat grains. The present work demonstrated isolation and cloning the promoter region of phytase gene. The detail analysis of amplified region revealed specific motifs which are mainly responsible for the regulation of phytase gene. PCR gene specific primers were synthesized to amplify promoter from C-306 and DBW 17 Indian wheat varieties. Different motifs viz., TATA box, CAAT box, GC box and other regulatory elements control the expression of promoter. Plant cis acting regulatory DNA elements database showed that seed specific motifs get activate during the development of seed. Seed-specific promoter was constituted with AAAG motif, ACGT motif and GCN4-like motif. Activity of phytase enzyme is primarily controlled at transcriptional level. Therefore, current research work could be more helpful to understand molecular mechanism which is mainly control the level of phytase activity.
