Summary Intraluminal vesicles accumulate within the endosomal lumen before lysosomal delivery or extracellular release. A new study reports the development of an elegant assay showing that these vesicles can escape from the endosomal lumen by ‘back-fusion’ or ‘retrofusion’ with the endosomal limiting membrane.
