Pharmacokinetic Study of Loganin in Fructus Corni in Rats

Objective:To establish a revered phase HPLC method for the study of pharmacokinetics of loganin in Fructus Corni after a single oral administration.Methods:The Hypersil C_(18)(4.6 mm×250 mm,5 μm) was used as the analytical column and the mobile phase consisted of acetonitrile-methanol-0.1% phosphoric acid(10∶5∶85) with the flow rate at 1.0 mL·min~(-1).The UV detection was set at 236 nm.The pharmacokinetic parameters of loganin were recorded.Results:After orally taken Fructus Corni,the plasma concentration-time course in rats fitted well to one-compartment model with the 1'st order absorption and with the following pharmacokinetic parameters:Ka=0.06 min~(-1),Ke=0.013 min,T(peak)=33.90 min,t_(1/2ka)=12.47 min,t_(1/2ke)=52.45 min.Conclusions:The HPLC method to determine plasma concentration of loganin and pharamacokinetic parameters in rats is feasible.
