Research on bronchial asthma-related cytokine regulation by spleen aminopeptidase

Objective To study the spleen aminopeptidase regulatory role for IL-10,IL-5 and TNF-α production and modulatory mechanism for IL-10. Methods CD4+ T cells and dendritic cells(DC) were isolated from human peripheral blood and co-cultured with chicken ovalbumin(OVA)as antigen to activate T cells,supernatants were collected after 48 hours,cytokines were detected by enzyme-linked immunoassay kit,and also investigated PI3K pathway regulating function on IL-10 using PI3K inhibitors. Results The trace amounts of IL-10,IL-5 and TNF-α secretion were detected in without OVA group, IL-10,IL-5,and the secretion of TNF-α were detected after CD4+ T cells and DC co-culture in joining OVA group,and the spleen aminopeptidase can promote the secretion of IL-10,but play an inhibitory effect on IL-5 and TNF-αsecretion.PI3K signaling pathway plays an important role in the secretion of IL-10. Conclusions Spleen aminopeptidase can promote in IL-10 secretion in vitro,inhibit the secretion of IL-5 and TNF-α,PI3K pathway plays an important role in the secretion of IL-10.
