Effects of Low-energy N+ Ion Implantation on Ethanol Fermentation Efficiency of Saccharomyces cerevisiae

2014
[Objective] To study effects of low-energy N+ion implantation on ethanol fermentation efficiency of Saccharomyces cerevisiae.[Method]S. cerevisiae AS2.399 was selected as recipient bacterium, the optimum implantation parameter was determined, effects of ion implantation on S. cerevisiae AS 2. 399 ethanol fermentation efficiency and key enzyme expression were studied. [Result] Under the condition of implantation dose of 10 × 1015ions /cm2,the survival rate of S. cerevisiae AS 2.399 was about 25%,ethanol yield higher than that of original strain was 0. 42 g ethanol /g glucose(84% of theoretical yield),and enzyme activity values of pyruvate decarboxylase and alcohol dehydrogenase relevant to ethanol formation which was also higher than thatof original strain was approximately 0. 53 and 2. 47,respectively. [Conclusion]This report could lay significant foundation for using low-energy ion implantation technology to recombine S. cerevisiae to expand its breadth of substrate utilization and improving its efficiency of ethanol fermentation.
