Changes of CD_4~+CD_(25)~+CD_(127)-regulatory T cells level and clinical significance in the peripheral blood of patients with lung cancer

2010
Objective To detect the levels of CD+4CD+25CD127-regulatory T(CD+4CD+25CD127-Treg) cells and the other related immunocyte subsets in the peripheral blood and plasma iterleukin-10(IL-10),interferon-γ(IFN-γ) levels in patients with lung cancer, and to evaluate the dysfunction of cell immunity and its clinical significance. Methods The levels of CD+4CD+25CD127-Treg cells and the other related immunocyte subsets in the peripheral blood were detected by flow cytometry(FCM) method, and plasma IL-10 and IFN-γ levels were determined by enzyme linked immunosorbent assay(ELISA) kits in 45 patients with primary lung cancer,14 patients with benign lung disease and 20 healthy volunteers. Results The levels of CD+4CD+25CD127-Treg cells and CD+4CD28-T cells in the peripheral blood of patients with lung cancer were significantly higher than those of patients with benign lung disease and healthy volunteers respectively. The levels of CD+4,CD+8CD+28-T cells,CD+56 NK cells and ratio of CD+4/CD+8 in the peripheral blood of patients with lung cancer were significantly lower than those in other groups.Plasma IL-10 level in patients with lung cancer was significantly higher than that in other groups.Plasma IFN-γ level in patients with lung cancer was significantly lower than that in other groups. The level of CD+4CD+25CD127-Treg cells in the peripheral blood was positively correlated with plasma IL-10 level and negatively correlated with plasma IFN-γ level in patients with lung cancer. The levels of CD+8CD+28,CD+56 NK and CD+4CD+25CD127-Treg cells, plasma IL-10 and IFN-γ levels were correlated with clinical stage. Conclusion The levels of CD+4CD+25CD127-Treg cells in the peripheral blood might play an important role in the development, progression of lung cancer.
