Effect of Chinese herbal medicine of removing toxic and blood stasis and nourishing yin on lymphocyte apoptosis in spleen of lupus mouse and expression of Cyt-C and Bcl-2.

Objective:To discuss the effect ofChinese herbal medicine ofremoving toxic and blood stasis and nourishing yin on lymphocyte apoptosis in spleen ofMRL/lpr mouse and expression ofCyt-C and Bcl-2.Methods:50 2-month-old male MRL/lpr mouse and 10 2-month-old male C57BL/6 mouse were randomly divided into six groups:high, middle and low dosage groups, pre-treatment group, model group and control group.After 30days treatment, the lymphocyte apoptosis in spleen oflupus mouse and expression ofCyt-C mRNA and Bcl-2 mRNA were detected.Results:The percentages oflymphocyte apoptosis in model group were(57.18±1.72)% lower than that in normal group(78.99±3.76)%(P0.01).The expression in spleen ofCyt-C mRNA was weakened and the expression ofBcl-2 mRNA was enhanced.With the early treatment ofChinese herbal medicine ofremoving toxic and blood stasis and nourishing yin, the percentages oflymphocyte apoptosis increased(P0.01).The expression ofCyt-C mRNA increased and the expression ofBcl-2 mRNA decreased(P0.05).Conclusion:The early treatment ofthe Chinese herbal medicine ofremoving toxic and blood stasis and nourishing yin can obviously enhance the percentages oflymphocyte apoptosis ofMRL/lpr mouse and the expression ofCyt-C mRNA .It can weaken the expression ofBcl-2 mRNA also.
