Quantitative Analysis of Urinary 11-Deoxy-17-ketosteroids and Pregnanediol by Gas-Liquid Chromatography

ABSTRACT A quantitative gas chromatographic method for the routine determination of urinary 11-deoxy-17-ketosteroids as their trimethylsilyl ethers, and of pregnanediol as the diacetate on a single column with 1 % SE-30 and 2% XE-60 as stationary phase, has been described. After hydrolysis, the 17-ketosteroids are purified by alumina column chromatography, converted into the trimethylsilyl ether derivatives, and quantitated by gas-liquid chromatography. Two procedures are described for the analysis of pregnanediol, using alumina column chromatography or Girard T separation as preliminary purification. Final purification is achieved by thin-layer chromatography (TLC) of the pregnanediol diacetate, followed by gas-liquid chromatography (GLC). The method has been applied to study the urinary excretion of androsterone, etiocholanolone and dehydroepiandrosterone in normal subjects before and after administration of corticotropin, Metopirone and dexamethasone.
