A Rapid and Efficient Method for Isolating High-Quality Total Proteins from Mature Buds and Other Woody Tissues of Vitis Vinifera

Protocols from most published plant proteomics studies use Trichloroacetic acid (TCA), acetone and phenols to extract and precipitate the proteins. However, these protocols are generally less effective for protein extraction from woody grapes tissues. The major challenges to protein extraction from these tissues include, among others, the accumulation of high levels of proteases and secondary metabolites , which interfere with the efficient extraction and proper separation of proteins. This section presents a fast and easy protocol for the extraction of total proteins from woody tissues of grapes.
