Rotavirus reassortant and its preparation

PURPOSE: To obtain a rotavirus reassortant useful for producing human rotaviral disease vaccines by simultaneous infection of a human rotavirus parent strain with an animal rotavirus to form a human rotavirus/animal rotavirus reassortant to which a specific antibody of an animal rotavirus glycoprotein is then subjected. CONSTITUTION: This rotavirus reassortant useful for producing vaccine precursor and vaccines against human rotavirus diseases is obtained by the following process: first, a human rotavirus parent strain incapable of culture is subjected to simultaneous infection with an animal rotavirus parent strain to afford a parent strain of human rotavirus/animal rotavirus reassortant; secondly, this parent strain is neutralized with a monoclonal antibody or hyperimmune guinea pig antisera which is specifically bound to the 38 kd glycoprotein of the animal rotavirus and neutralizing the animal rotavirus parent strain; finally, the resultant parent strain so neutralized is purified, via a plaque, into such a form as to be suitable to vaccines to prepare the aimed reassortant containing both human and animal genes and having human rotavirus antigenic serum type.
