Effects of Rosiglitazone on Neointimal Formation of Balloon Injured Rat and Its Mechanism

【Objective】To investigate the effects of rosiglitazone(RSG) on neointimal formation after balloon injury and its mechanism.【Methods】 Endothelial denudation of aorta in SD rats was performed with 2F Fogarty balloon catheter.Male rats were randomly divided into sham-operated group,control group,and RSG group.Aortic tissues were harvested in 14th day after injury.The endothelial regeneration rate,IA/MA ratio,P27 and PCNA expression index,and cell cycles were detected.【Results】 Re-endothelialization of the injured aorta in RSG group was enhanced as determined by Evans blue stain(P0.01).There was a significant decrease of the neointima in RSG groups(P0.01).The expression of P27 was greater and that of PCNA was lower in RSG group than that in control group(P0.01).The Flow Cytometry showed that the percentage of S and G2/M phase cells in RSG group were lower than that in control group.【Conclusion】 RSG can obviously attenuate neointimal formation of the aorta after balloon injury and its mechanism may be related to up-regulation of P27 and inhibition of cell cycle progression.Otherwise RSG may also inhibit proliferation of the intima by promoting endothelial regeneration.
