Induction and differentiation of protocorms of Dendrobium nobile and Pleion maculate on different media

Dry seeds of Pleion maculate and Dendrobium nobile were cultured on MS,B5 and KC liquid media for inducing protocorms,and the protocorms were subcultured on MS,B5 and KC solid media for observing protocorm differentiation.The experimental results showed that protocorms from the orchid seeds of two species cultured on B5 medium were most.Protocorms of Pleion maculate on KC medium were of the highest viability,and developed best.Protocorms of Dendrobium nobile on B5 medium were of the highest viability and developed best on KC medium.The present study suggested that orchid seedling in different stages of development required different nutrients.Selection of optimum media for orchids in different culture stages is crucial to obtain excellent plant strains and establish clones of high productivity.
