Establishment and evaluation of combining disease and syndrome model of non-alcoholic fatty liver disease with syndrome of blood stasis

Objective: To establish a model combining disease and syndrome of non-alcoholic fatty liver disease with syndrome of blood stasis.Methods: 76 rats were randomly divided into normal group of 10,NAFLD group of 30 and blood stasis NAFLD group of 36.The rats in normal group were fed with normal diet,other groups were fed with high-fat diet.Simultaneously,the rats in the blood stasis NAFLD group were dealt with norepinephrine(NE) and bull serum albumin(BSA).At the end of 4th,8th and 12th weeks,the rats were disposed.Physical signs were observed,skin micro-circulation was detected with DRT4 laser Doppler blood stream tester.Hemarheology was tested with automatic blood rheology detector,serum lipidsa and serum ALT and AST were examined with automatic biochemistry analyser,fatty degeneration and inflammation of liver were observed with oil red O and HE dyeing.Results: The rats of blood stasis NAFLD group showed sublingual varicose veins,cyanoderma tongue with ecchymosis,black red eye ball,tail ecchymosis,skin micro-circulatory disturbance,abnormity of hemarheology,disorder of serum lipids and fatty degeneration and inflammatory cell infiltration.The integration of blood stasis physical signs and whole blood viscosity,whole blood reduction viscosity and RBC aggregation index in the blood stasis NAFLD group were higher than those in the NAFLD group.Conclusion: Blood stasis NAFLD model can be successfully induced by high-fat diet,NE and BAS.This model has either the characteristic of pathology of NAFLD,or the syndrome performance of blood stasis of TCM.
