MDM2 modulators of proteolysis and related methods of use

The present description relates to the imide-based compounds comprising a bifunctional compound composed of the MDM2 binding compound, and these compounds are particularly useful as modifiers of targeted ubiquitination, particularly for the bifunctional compounds of the present invention. It finds utility as an inhibitor of various polypeptides and other proteins that are degraded or otherwise inhibited by compounds. In particular, the present description contains a ligand that binds to MDM2 E3 ubiquitin ligase at one end and binds to the target protein so that the target protein is placed in the vicinity of the ubiquitin ligase and degradation (and inhibition) of the protein occurs. Compounds are provided that contain a moiety at the other end. Compounds can be synthesized that exhibit a wide range of pharmacological activities consistent with degradation / inhibition of almost any type of target polypeptide.
