Effects of Citicoline, Meclofenoxate, GM1 Ganglioside on Some Intracerebral Endogenous Substances in Rats with Epilepsy Induced by Penicillin

Objective To study the effects of citicoline, meclofenoxate, GM1 ganglioside on the some itracerebral endogenous substances in the rats with epilepsy induced by penicillin. Methods Thirty-five SD rats were randomly divided into five groups: control group,model group, citicoline group, meclofenoxate group and GM1 ganglioside group. The model of epilepsy was made by intraperitoneal injection of penicillin. One hour after the injection of penicicllin, the animals were sacrificed and the contents of superoxide dismutase(SOD), malonaldehyde(MDA), glutamate(Glu), gamma-aminobutyric acid(GABA) in the frontoparietal cortexes were determined in all the rats. Results ① The models of epilepsy were successfully made. Pretreatments with citicoline, meclofenoxate, GM1 ganglioside could not prevent the development of the seizure in the rats receiving the injection of penicillin. ② Compared with the control group, the SOD activity decreased significantly (P0.01), the level of MDA, Glu and the ratio of Glu to GABA increased signi-ficantly (P0.05) in cerebral cortex in the model group. ③ Compared with the model group, the SOD activity increased significantly (P0.05), the level of MDA and the ratio of Glu to GABA decreased significantly (P0.05) in citicoline group, meclofenoxate group and GM1 ganglioside group, and the contents of Glu decreased significantly only in GM1 ganglioside group. Conclusions The pretreatments with citicoline, meclofenoxate, GM1 ganglioside can not prevent the development of the seizure in the rats treated by penicillin. But they have protective effects on the cerebral tissues by scavenging free radicals, increasing antioxidation, decreasing the ratio of Glu to GABA, balancing neurotransmitter and relieving excitotoxiaty in the rats with epilepsy induced by penicvillin.
