Reversal effect of Wogonin on cisplatin-resistance of human ovarian cancer cells

2016
Objective: To investigate the reversal effect of Wogonin on cisplatin (DDP)-resistance of human ovarian cancer SKOV3/DDP cells, and to explore its possible mechanism. Methods: SKOV3/DDP cells were treated with 20 µmol/L Wogonin for 24 h, while the SKOV3 cells and the SKOV3/DDP cells without any treatment were used as the controls. The cell proliferation ability was detected by CCK-8 method. The cell migration and invasion abilities were detected by scratch wound healing and Transwell invasion assay, respectively. Furthermore, the expressions of P-glycoprotein (P-gp), E-cadherin, N-cadherin and vimentin proteins were detected by Western blotting. Results: After treatment with Wogonin, the sensitivity of SKOV3/DDP cells to DDP was increased (P 0.05).Conclusion: Wogonin may increase the sensitivity of SKOV3/DDP cells to DDP by reversing epithelial-mesenchymal transition.
