Cloning of Cold-regulated Transcrptional Factor CBF2 and Construction of Plant Expression Vector

C-repeat binding factor 2 and hypothermal inducer promoter Rd29A were amplified from the genomic DNA of Arabidopsis thaliana Columbia.Sequencing results showed that the cloned CBF2 gene had 725 bps and encodes 216 amino acids,and had 99.8 % identity with the sequence published on GeneBank;the promoter Rd29A had 950 bps,and had 99.5 % identity with the already published promoter sequence.Based on the binary vector pCAMBIA1301,we successfully constructed the plant expression vector pCAMBIA1301-Rd29A-CBF2,which could materially improve the cold-tolerance ability of plants.
