The invention discloses an aeromonas hydrophila detection method comprising the following steps: preparing rabbit anti-aeromonas hydrophila serum; purifying the rabbit anti-aeromonas hydrophila serum by using a SPA affinity chromatography column to obtain rabbit anti-IgG; preparing biotin labeling rabbit anti-IgG; carrying out time resolved fluorescence (TRF) immunoassay on biotin-streptavidin; and replacing bacterium suspension with assay buffer to serve as negative control, comparing with TRF, and when the ratio of the TRF value of a sample hole to the TRF value of a control hole is larger than 2, judging a positive result, or when the ratio is smaller than 2, judging a negative result. On the basis of controlling the dissociation time to eliminate granular antigen interference, a biotin-streptavidin signal amplification system is introduced to detect the aeromonas hydrophila, and the sensitivity reaches 1.0*102 cfu/mL. Meanwhile, the TRF and bacteria concentration logarithm form a linear relation, and quantitative detection of the aeromonas hydrophila can be carried out.
