Development of specific antibodies against major structural proteins of Newcastle disease virus Class I virulent strain.

To investigate each protein's function,antisera and monoclonal antibodies(McAbs) against NP,P,M,HN,and F proteins of Newcastle disease virus(NDV) were developed with ClassⅠ 9a5b virulent strain.Hemagglutination inhibition,indirect immunofluorescence assay,cell neutralization test,ELISA and Western-blot analysis were used to characterize and identify these antibodies.The results showed that,besides antisera against each protein,a panel of 12 hybridoma cell lines were successfully prepared.Among them,McAbs 3H7 and 3H9 were specific to HN protein with the ability of neutralizing NDV and inhibiting hemagglutination.McAb 2A8 was another ClassⅠ NDV specific antibody except 3H7 and 3H9.All the other McAbs specific to P or NP protein had a cross-reaction with ClassⅡ NDV strains.Therefore,specific antibodies were purified and accumulated for further studies on pathogenesis of ClassⅠ NDV.
