Concentrations of Endothelial Progenitor Cells CD34+ and Interleukin-8 in Patients With Different Types of Coronary Artery Disease and Its Significance

Objective: To investigate the concentrations of endothelial progenitor cells(EPCs) CD34+ and interleukin-8(IL-8) in patients with different stenotic types of coronary artery disease and its significance.Methods: One hundred and thirty-two cases confirmed as coronary artery disease(CAD) by coronary angiography were selected.Patients were divided into 3 groups according to their lesion characteristics: type A,B and C group,meanwhile,40 healthy volunteers severed as control group.Mononuclear cells were isolated by density-gradient centrifugation from all the patients and control group.Flow cytometry was used to identify EPCs.In addition,dual-stained positive cells were counted.EPCs adhesive,migratory and vasculogenesis capacity were assayed by adhesion assay,modified Boyden chamber and In Vitro Angiogenesis Assay. Serum IL-8 was measured by ELISA.Results: ① Concentration of serum EPCs CD34+ was significantly lower in all types of CAD group than that in control group.With the aggravating of coronary lesions,concentration of serum EPCs CD34+ showed a decreasing tendency.Concentration of serum EPCs CD34+ in type C group was significantly lower than that in type A and B group,and that in type B was significantly lower than that in type A(P0.05).② With the aggravating of coronary lesions,the numbers of dual-stained positive cells,adhesive cells,migratory cells and vasculogenesis capacity cells were descend gradually.③ Concentration of serum IL-8 in all CAD groups was significantly lower than that in control group.Concentration of serum IL-8 in type C was significantly higher than that in type B(P0.05).④ Serum EPCs CD34+ and IL-8 was negatively correlated in patients with CAD(r=0.672,P=0.009).Conclusions: Concentrations of EPCs CD34+ and IL-8 were closely correlated with types of coronary lesions.EPCs CD34+ and IL-8 were negatively correlated.Decreased number of EPCs CD34+ and its poor function are closely correlated with the attack of CAD and its severity.
