Construction and evaluation of full-length cDNA plasmid library from adult Echinococcus granulosus.

Objective: To construct and evaluate full-Length cDNA library of adult Echinococcus granulosus(E.g.).Methods:mRNA was extracted from the adult E.g.and used for the construction of full-length cDNA library using SMART-pBluescriptⅡSK kit.The recombination rate and capability of the library was measured and the length of insert fraction of the positive recombinant clones was tested by PCR.Positive recombinant clones were randomly selected for 5'end sequencing and the rates of unigene,full-length cDNA was analyzed by EST data using bioinformatics.Results: The full-length cDNA libray of adult E.g.was constructed successfully.The recombination rate and capability of the library were 95.04% and 1.13×106,respectively.The average length of insert fraction was about 1.2kb.24 recombinant clones randomly selected were sequenced,the rate of unigene and full-length cDNA were 69.57% and 64.71%.Conclusions: A high-quality of full-length cDNA plasmid library of adult E.g.has been constructed successfully.
