Effect of HE4 Overexpression on the Proliferation and Invasion Abilities of the Ovarian Cancer Cell Line HO8910

2009
Objective To examine the effect of HE4 gene overexpression on the proliferation and invasiveness of the ovarian cancer cell line HO8910. Method HE4 carrying PcDNA3.1 vector(PcDNA3.1 / HE4)was constructed and transfected into a highly metastatic human ovarian cancer cell line HO8910 with Lipofectamine 2000. The protein expression was determined by Western-blot. MTT and colony formation assay were used for cell growth measurement. Tumour cell invasiveness and cell adhesiveness were also determined. Result Overexpression of HE4 was confirmed by Western-blot and the growth was not affected by HE4 overexpression. However, the tumour cell invasiveness was reduced. Conclusion Overexpression of HE4 can reduce the invasiveness of HO8910 cells, and HE4 may be a candidate gene for the treatment of ovarian cancer.
