Studies on the Distribution and Properties of Scylla serrata Lectins

Lectin of Scylla serrata was studied by biochemistry methods.The results showed that lectins distributed not only in the muscle but also in the serum of Scylla serrata.There were lots of different properties between these two lectins, such as thermostability,the range of pH and the dependence on metal ionic.Muscular extract agglutination quail erythrocytes depended on Ca~(2+) evidently,and it keep 100% agglutinative activity in the range of pH 5.0~8.0,it had agglutinative activity in temperature range of 4~90℃.Agglutinative activity of serum to quail erythrocytes also depended on Ca~(2+),but it still had agglutinative activity when EDTA had added up to 50 mmol/L,it revealed that the degree of depending on Ca~(2+) is small compare with muscular extract.And it had higher agglutinative activity in the range of pH 7.0~9.0,its agglutinative activity lost at 80℃ by 10 minutes.Adding solid ammonium sulfate into the muscles and serums up to 50% saturation at 4 ℃,the precipitates were continually extracted by ammonium sulfate with different concentration.The extractions from muscle and serum given a 2.25-fold and 1.27-fold purification individually,(33.36%) and 16.70% recovery of total activities compared with the muscle and serum.
