Steady-state optical pumping up-conversion luminescence flow cytometry detection device
2011
The invention belongs to the technical field of biological detection, and concretely relates to a novel steady-state optical pumping up-conversion luminescence flow cytometry detection device. The device comprises a steady-state light source, a focusing lens, a sample flow chamber, an up-conversion luminescence collection lens, light filters and a detector unit. The device is characterized in that steady-state light beams generated by the steady-state light source having the center wavelength in a range of 500-1500nm are introduced to a flow cytometry detection system through fibers, the focusing lens is placed along the forward direction of the steady-state light beams to form an oval light spot with the geometric dimension being slightly greater than a cell diameter, and the steady-state light beams are orthogonal to the sample flow in the flow chamber; and the up-conversion luminescence collection lens, the light filters with the wavelengths of less than the wavelength of the light source, and detectors are sequentially arranged in the orthogonal direction traversing the sample flow and the steady-state beams. The device can be used for direct light intensity analysis and sorting of materials (with the particle size being greater than 500nm) having the steady-state optical pumping up-conversion luminescence property, and can also be used for qualitative and quantitative analysis and detection of living cells or other matrixes containing up-conversion luminescence particles.
