Early Inhibition of Glycosylation with Deoxynojirimycin Has No Effect on the Replication of Flavivirus West Nile

Deoxynojirimycin (dNM) is a specificandreversibleinhibitorof the trimmingac- tions ofglucosidaseI and II in the roughendoplasmic reticulum. Theimportance of the ef- fects of dNMon the replicationof West Nile (sarafend- glycosylated) and West Nile (Wengler- nonglycosylated) viruseswere studied.It was foundthat dNMtreatmentin- creasethe molecular weightsof the E andprMproteinsin bothvirusstrainswithoutaffec- ting the yieldsof the virus. The intracellulartransportof the E proteinfromthe roughen- doplasmicreticulumto Golgiapparatuswas not affecteddespitethe changein molecular weights.It wouldappearthat the preventionof removalof glucosemolecules by dNMdid not affectedthe maturationof E proteinor the virus.
