Experimental Study the on Relationship between Inhibiting the Ubiquitin-Proteasome Pathway and Drug Sensitivity in Human Liver Carcinoma Cell

目的 研究阻断泛素-蛋白酶体通路与人肝癌细胞药物敏感性的关系及可能机制.方法 以不同浓度的特异性泛素-蛋白酶体抑制剂MG-132处理人肝癌细胞株HepG2作为实验组,化疗药物为氟尿嘧啶、表阿霉素、丝裂霉素.MTT法检测细胞的代谢活性(IC50值),流式细胞术和Hoechst荧光染色检测细胞凋亡,半定量逆转录-聚合酶链反应(RT-PCR)和Western blot检测caspase-3基因的转录水平和蛋白水平.结果 实验组的IC50值明显低于对照组,细胞凋亡率、caspase-3的转录水平及蛋白水平明显高于对照组.结论 阻断泛素-蛋白酶体通路能增加人肝癌细胞的药物敏感性,其机制可能与caspase-3基因的表达水平升高有关。
