The invention discloses a multi-chamber microfluidic PCR chip for screening aptamer. The multi-chamber microfluidic PCR chip comprises a micro-flow PCR reaction piece, a temperature circulating heating plate, a glass substrate and a chip fixture, wherein a reaction liquid filling hole, a Z-type mixing unit, an inflow micro-flow channel, a PCR amplification chamber, an outflow micro-flow channel and a reaction liquid outflow hole which are positioned on a PDMS reaction plate are sequentially communicated with one other. According to the microfluidic PCR chip, a plurality of reaction chambers are adopted, so that the high quality and sufficient acquisition of a secondary library and the variety of nucleic acids can be guaranteed; an optimal amplification condition of the microfluidic PCR chip can be quickly acquired by fluorescent real-time amplification curves, and nucleic acid sequences screened by aptamers are amplified under an optimal condition. The invention also discloses a method for screening the aptamers by utilizing the multi-chamber microfluidic PCR chip; the method can be used for PCR amplification for screening various types of target aptamers and provides a good thought design and a reference for other aptamer screening workers.
