The invention discloses a pseudomonas aeruginosa antibody detecting kit which is prepared from an enzyme-labeled plate, pseudomonas aeruginosa outer membrane protein wrapped by an enzyme-labeled plate solid-phase carrier, positive serum, negative serum, a scrubbing solution, a color-substrate solution, an enzyme-labeled second antibody and a stop solution. Meanwhile, the invention further provides a detecting method. The method includes the steps of adding diluted to-be-detected serum into holes of the enzyme-labeled plate, adding the scrubbing solution for standing and washing after the reaction is completed, adding the diluted enzyme-labeled second antibody into the enzyme-labeled plate to act for 0.5-3 h at the temperature of 37 DEG C, adding the scrubbing solution, standing for 3-10 min, conducting washing, adding the color-substrate solution for a reaction for 10-20 min after washing is completed, adding the stop solution for stop after a chromogenic reaction is completed, and detecting the readings at the wavelength of 450 nm and the reference wavelength of 630 nm through an enzyme-labeled instrument. By means of the detecting kit and method, the antibody in musk deer serum can be detected efficiently and rapidly, high specificity is achieved, operation is easy, and cost is low.
