Disclosed are a macromolecular intravital staining contrast media, and a preparation method and use thereof. The contrast media uses a polyuronic acid as a carrier, a mannose or mannose derivative as a mannose receptor (MBP) recognition group, and a paramagnetic metal ion chelate as a nuclear magnetic resonance imaging group, which is used for the sentinel lymph node location before operating on breast cancer, head and neck cancer, melanomas and so on, and satisfies the biopsy and nuclear magnetic resonance imaging, and is a bifunctional lymphatic targeted contrast media.
