The Stereological Study on Pyramidal Cells and Astrocytes in Rat Hippocampus after Cerebral Ischemia-Reperfusion

To investigate the changes of pyramidal cells and astrocytes in rat hippocampus after ischemia-reperfusion,24 SD rats were randomly divided into four groups which were control group,ischemia-reperfusion 6 h group,12 h group and 24 h group.Changes in hippocampus CA1,CA3 were observed by Nissl staining and anti-GFAP immunohistochemistry,stereological method was used to evaluate changes as well.The results showed that the volumn density(Vv) and surface density(Sv) of pyramidal cell in CA1 and CA3 decreased as reperfusion time went on,and reperfusion 6 h,12 h,24 h groups had distinct differences compared with control group(p0.05,p0.01).While the Vv and Sv of astrocytes increased with the reperfusion time,compared with control group,6h group had no significant difference(P0.05),12 h,24 h group had significant difference(p0.01,p0.01).There were no distinct differences between CA1 and CA3 of the same group as well as between the left and right side of the same region(p0.05).These results indicated that neuron damage became more severe as reperfusion time extended,while astrocytes functioned more actively and were closely related with neurons damage degree.
