Study on long noncoding RNA in urothelial carcinoma of bladder

Objective To compare the gene expression differences between human urothelial carcinoma and normal bladder tissues,and the significances in utothelial carcinoma of bladder.Methods We collected 12 specimens of urothelial carcinoma and 3 specimens of normal bladder mucosa,then we extracted RNA,transcribe it into cDNA and labeled it.Then we used the probe which have already be labeled hybridize with microarray.After washing,slides were scanned with the Agilent DNA Microarray Scanner.Data was extracted using Agilent Feature Extraction software.Results Compared with nomarl bladder tissues,we found 7 differentially expressed genes by Enhancer long noncoding RNA (LncRNA) profiling and 25 differentially expressed genes by Rinn LncRNAs profiling.Then we used reverse transcriptase-polymerase chain reaction (RT-PCR) assay to amplify three of the differentially expressed genes,its expression consistent with the microarray results.Conclusion LncRNA have close relationship with urothelial carcinoma' s developing.It may be an significant tumor marker for urothelial carcinoma of bladder,which can help us make early diagnosis,prediction and open new paths of gene therapy for bladder urothelial carcinoma.
