Summary We haverecentlyshown thatCD30, amember ofthetumor necrosisfactor/nerve growthfactorreceptorsuperfamily, ispreferentially expressedby human T cellclonesproducingT helper (Th)type2 cytokines .We reportherethatcostimulation withan agonistic anti-CD30monoclonalantibodyenhancedantigen(Ag)-inducedproliferation and cytokinesecretionby establishedhuman Th2 andTh0 clones.Moreover,costimulation ofperipheral bloodmononuclear cellswiththesame anti-CD30 monoclonalantibodyresultedinthepreferential development ofAg-specific T celllinesandclonesshowingaTh2-likeprofileofcytokinesecretion .Incontrast, earlyblockadeinbulkcultureofCD301igand-CD30 interaction shiftedthedevelopment ofAg-specificT cellstowardstheopposite(Th1-like)phenotype.Taken together,thesedata suggestthatCD30 triggering ofactivated Th cellsbyCD30 ligand-expressing Ag-presentingcells may representanimportantcostimulatory signaling forthedevelopmentofTh2-typeresponses .
