Screen of nicotine-associated protein induced by JA simulated flue-cured tobacco topping

To study the molecular mechanism of nicotine synthesis stimulated by topping and the action of JA Signaling pathways on it.We simulated flue-cured tobacco topping with JA.The proteomic of flue-cured tobacco root tip were analyzed by technology and method of differential proteomics.The results analyzed by Image Master 2 D software showed that there were about 500 detectable spots which were separated distinctly.By comparison,the up proteins in topping and JA treatement were thirteen and twenty-six,and there were three common spots.The down proteins were five and six,and there were three common spots.Eight and five differential expressed spots were found respectively,and there were four common spots.With direct MALDI-TOF mass spectrometry analysis and protein database searching,eight protein spots were identified,in which four differential proteins showed the same trend in topping and JA treatment,suggesting that they act as nicotin-regulated biosynthesis.
