The significance of CRT_(H)2 expression on T cell subsets with idiopathic thrombocytopenic purpura

To determine CRT_(H)2 expression on T cell subsets in pediatric patients with idiopathic thrombocytopenic purpura (ITP). The ratios of CD4~(+)/CD8(+),CD4~(+)CRT_(H)2~(-)/CD4(+)CRT_(H)2~(+) and CD8~(+)CRT_(H)2~(-)/CD8~(+)CRT_(H)2~(+) in peripheral T cells were analyzed by immunofluorescince staining and bicolorflow cytometry (FCM). Compared with the ratios of CD4~(+)CRT_(H)2~(-)/CD4~(+)CRT_(H)2~(+) (47.21%±5.36%) and CD8~(+)CRT_(H)2~(-)/CD8~(+)CRT_(H)2~(+ )(20.47%±4.03%) in healthy children, the ratios of CD4~(+)CRT_(H)2~(-)/CD4~(+)CRT_(H)2~(+)(61.18%±7.34%) and CD8~(+)CRT_(H)2~(-)/CD8~(+)CRT_(H)2~(+) (25.06%±3.28%) in ITP children significantly increased. FCM analysis showed that the average percentages of CD4~(+),CD4~(+)CRT_(H)2~(-), CD4~(+)CRT_(H)2~(+ ),CD8~(+),CD8~(+)CRT_(H)2~(-) and CD8~(+)CRT_(H)2~(+) were 21.82%±6.5%, 21.48%±6.3%, 0.34%±0.16%, 33.14%±6.15%, 31.83%±5.48% and 1.47%±0.97% in ITP children versus 39.03%±5.6%, 38.24%±5.5%, 0.81%±0.19%, 33.06%±5.63%, 31.52%±5.19% and 1.51%±0.68% in healthy children. The average percentages of CD4~(+),CD4~(+)CRT_(H)2~(-),and CD4~(+) CRT_(H)2~(+) obviously decreased in children with ITP. At the same time, the average percentages of CD8~(+),CD8~(+)CRT_(H)2~(-) and CD8~(+)CRT_(H)2~(+) didnot changed. The ratios of CD4~(+)CRT_(H)2~(-)/CD4~(+)CRT_(H)2~(+) and CD8~(+)CRT_(H)2~(-)/CD8~(+)CRT_(H)2~(+) in peripheral T cells increased in ITP children. It suggested that cellular immunity in ITP children is shifted to T_(H)1 type immunity, which may play an important role in the pathogenesis of ITP.
