Effects of Sericine on HO-1 Expression in Hippocampus of Type 2 Diabetic Rats

Objective To investigate the effects of sericine on heme oxygenase-1(HO-1)expression in hippocampus of type 2 diabetic rats.Methods 30 male SD rats were randomly divided into 3 groups(n =10 respectively):normal control group,diabetes model group and sericine treatment group.The diabetes model rats were made by continuous intraperitoneal injection of streptozotocin and blood glucose≥ 16.7mmol/L was taken as the standard of model-formation.After successfully establishing diabetes rat model,rats in diabetes model group were not treated anymore;rats in sericine treatment group were lavaged with sericine(2.4 g·kg-1·d-1)for 35 days.The morphological changes of nerve cells in hippocampus were observed by HE staining.Western Blotting and RT-PCR were respectively used to detect the expression of HO-1 protein and mRNA in hippocampus.Results In diabetes model rats,the neurons in CA1 region of hippocampus showed obvious pathological changes,the expressions of HO-1 protein and mRNA were obviously higher than normal control rats(P 0.01).Compared with diabetes model rats,the pathological changes of neurons were alleviated significantly(P 0.01),the expressions of HO-1 protein and mRNA in CA1 region of hippocampus in sericine-treated rats decreased significantly(P 0.05).Conclusion Sericine can alleviated the damages of neurons in hippocampus induced by high expression of HO-1,thus has protective effects on injuries of nervous system during diabetes.
