The work deals with the determination of cholinesterases activity in human blood. Cholinesterases are enzymes which preferentially hydrolyse choline esters and perform a variety of biological functions. There are two different types of cholinesterases in the blood acetylcholinesterase and butyrylcholinesterase. A part of this work is to determine the activity of these two enzymes in blood samples from volunteers using spectrophotometric Ellman's method and pH-stat method. The work also includes determination of inhibitory potency of selected salicylanilide thiophosphates against both cholinesterases in human blood. For this purpose were used Ellman's and pH-stat methods. It was also determined the partition coefficient for all tested salicylanilide thiophosphates in system 1-octanol:water. Finally the type of inhibition for the selected salicylanilide thiophosphate inhibitor using Ellman's and pH-stat metods was also defined. Obtained experimental results from both methods were compared with each other and there were observed differences between resulting values. This may be caused not only by using different methods of measurement, but also a detection of another product.
