Expression of Bacillus sp. N-4 CMCase Gene in Saccharomyces cerevisiae

A 5.5kb HindIII fragment of pYBC107 containing carboxymethyl cellulase (CMCase) gene from Bacillus sp. strain N-4 (ATCC 21833) was inserted into the HindIII site of YEp13. The reconstructed plasmid, named pYES450, was isolated from Escherichia coli transformants that produced CMCase. With the 16kb plasmid pYES450 Saccharomyces cerevisiae DBY-746 was transformed, and expression of the CMCase gene in the host was investigated. Plasmid pYES50 maintained in S.cerevisiae stably. The CMCase activity was low and was only detected in the cell extract of the S.cerevisiae transformant.
