The present invention discloses a method for preparing a surface-enhanced Raman scattering substrate. The silver nitrate is completely dissolved in pure water; then adding a cationic ion exchange resin reaction; was added after the reaction of the resin with pure water using an appropriate amount of water, which was placed in an ice bath, was added a solution of borohydride reduction the reaction, silver coated surface to obtain a surface-enhanced Raman scattering after washing and drying the substrate. Using a substrate prepared by the method, its spectral signal stability and good reproducibility, a long storage time base, and easy preparation, lower cost, and analysis can be used to detect low levels of the compound.
